I. Chemical Structure and Properties

Molecular Structure: The benzene ring is connected to a sulfonamide group (-SO₂NH₂), with the chemical formula C₆H₇NO₂S and a molecular weight of 173.2 g/mol.

Physical Properties:

It is a white crystalline powder, with a melting point of approximately 156–158℃. It is slightly soluble in water and readily soluble in organic solvents (such as ethanol and ether).

Chemical Properties:

The hydrogen atom in the sulfonamide group can be substituted to form N-substituted benzenesulfonamides (such as sulfonamide drugs). It has acidity (pKa is approximately 10–12) and can form salts with bases.

Benzenesulfonamide is the core framework of sulfonamide antibacterial drugs. By introducing different substituents (such as pyrimidinyl, thiazolyl, etc.) on the N atom, a series of drugs with antibacterial activity are formed. For example:

·Sulfadiazine: Used to treat bacterial infections (such as meningitis), with the N atom connected to the pyrimidine ring.

·Sulfamethoxazole: Used in combination with trimethoprim (cotrimoxazole) for respiratory and urinary tract infections.

·Mechanism of action: By inhibiting bacterial dihydrofolate synthetase, blocking folic acid metabolism, and inhibiting bacterial proliferation.
